Histology Sample | Preparation |
Duodenum | The small intestine is dissected and mesentery removed. The proximal 5 cm are discarded and the tissue is divided into thirds lengthwise. The duodenum (proximal 1/3) is flushed with sterile PBS, opened longitudinally, then swiss rolled from the distal end to the proximal. Tissues are fixed in 10% neutral buffered formalin (24hrs), transferred to 70% ethanol at 4˚C (<1 week). The tissue is then processed and paraffin-embedded. |
Jejunum | The small intestine is dissected and mesentery removed. The proximal 5 cm are discarded and the tissue is divided into thirds lengthwise. The jejunum (middle 1/3) is flushed with sterile PBS, opened longitudinally, then swiss rolled from the distal end to proximal. Tissues are fixed in 10% neutral buffered formalin (24hrs), transferred to 70% ethanol at 4˚C (<1 week). The tissue is then processed and paraffin-embedded. |
Ileum | The small intestine is dissected and the mesentery removed. The proximal 5 cm are discarded and the tissue is divided into thirds lengthwise. The ileum (distal 1/3) is flushed with sterile PBS, opened longitudinally, then swiss rolled from the distal end to proximal. Tissues are fixed in 10% neutral buffered formalin (24hrs), transferred to 70% ethanol at 4˚C (<1 week). The tissue is then processed and paraffin-embedded. |
Colon | The colon is dissected and the mesentery removed. The colon is flushed with sterile PBS, opened longitudinally, then swiss rolled from the distal end to proximal. Tissues are fixed in 10% neutral buffered formalin (24hrs), transferred to 70% ethanol at 4˚C (<1 week). The tissue is then processed and paraffin-embedded. |
